Problem is solved.

Turned out to be an issue with Remote Desktop. I am setting this computer up remotely, and for whatever reason, Voicemeeter can't work in a Remote Desktop environment.

When I tested locally, everything looked fine.

I've gone through this a few times now. New Win-10 install, on new computer build Motherboard has audio working before. I have Voicemeeter Potato licensed and installed.
